Niacinamide (or nicotinamide) is a type of vitamin b3, a pyridine derivative, that is found in many foodstuffs like vegetables, meat, cereals, and fruits. This non-irritant & water-soluble chemical is known for its brilliant ability to reduce blemishes and acne scars over time, give a more youthful appearance and fight against free radical damage. It also has a similar pH to that of our skin, i.e., 5.5-5.6.
ACNE FORMATION
Our skin has pores containing hair follicles and sebaceous glands (sebum-producing microscopic organs), which help seal moisture inside the skin and prevent foreign substances from entering. When dead skin cells are stuck together by keratin, they start layering on top of these pores, clogging them while the sebaceous gland keeps producing sebum, which gets trapped inside the clogged pores. These pores, when infected with bacteria, form acne or pimples.
HYPERPIGMENTATION
Amino acid tyrosine is oxidized by the enzyme tyrosinase to form melanin. Melanosomes are intracellular organelles, present in melanocytes (melanin-producing cells), that are responsible for producing and storing melanin. These melanosomes are transferred from melanocytes to keratinocytes because of which pigmentation occurs. However, during excessive UV exposure, hormonal changes, and aging tyrosinase activity increases, because of which the amount of melanin produced is more and melanosome transfer from melanocytes to keratinocytes also increases, which causes hyperpigmentation.
AGING
Skin cells are present in layers, the stratum corneum (outermost layer) has dead skin cells while the cells below the stratum corneum are still young and maturing. Furthermore, desmosomes are ‘binding bodies’ in animal tissue that anchor two adjacent cells together, keeping skin cells organized. During metabolism, enzymes break down desmosomes of dead skin cells, leading to the separation of dead skin cells and subsequent shedding of the stratum corneum. This process of dead skin cells shedding and the younger cells replacing them is called skin cell turnover. However, with aging the rate of metabolism decreases because of which the enzymes causing the breakage of desmosomes are produced at a very slow rate. In that case, the dead skin cells stay intact causing the build-up of dead skin layers, hence slower skin cell turnover.
STRUCTURAL INTEGRITY OF SKIN CELLS
During glycation, covalent bond forms between amino acids on collagen fibers and amino acids on elastin fibers. Because of the presence of covalent bonds, the bonds are difficult to break, this causes the skin to become rigid.
MECHANISM OF ACTION
NADH (Nicotinamide adenine dinucleotide + hydrogen) and NADPH (Nicotinamide adenine dinucleotide phosphate) are coenzymes that are like energy units in a cell. They carry forward metabolic activities in a cell, but with age NADH and NADPH, concentration decreases because of which the rate of metabolism decreases. Upon supplementing niacinamide topically, the concentration of NADH and NADPH increases, subsequently It also prevents the loss of water from the skin, keeping it hydrated.
Niacinamide is an anti-inflammatory and anti-microbial ingredient, so it prevents acne-forming bacteria from growing, eventually preventing acne formation. Also, it reduces sebaceous lipogenesis (sebum production), making it a good ingredient for oily skin.
Niacinamide regulates collagen production and reduces the rate of glycation, keeping the collagen framework intact. This helps in reducing wrinkles and fine lines, eventually making your skin look younger. Niacinamide also decreases UV-mediated damages, it decreases the number of free radicals generated when the skin is exposed to UV radiation.
